OR11-005 - Mast cells respond to pathogen signals with IL-1ß
نویسندگان
چکیده
Introduction Mast cells, key effector cells of allergic and innate immune responses, have recently been reported to be an important source of IL-1ß in patients with autoinflammatory conditions such as cryopyrin-associated-periodic-fever syndromes (CAPS). CAPS patients show IL-1beta-driven systemic inflammation together with non-histamine dependent urticarial rash, which are caused by activating mutations of the inflammasome, a multiprotein oligomer responsible for the initiation of inflammatory responses to pathogens.

Selective activation of Fyn/PI3K and p38 MAPK regulates IL-4 production in BMMC under nontoxic stress condition.
Mast cells have the ability to react to multiple stimuli, implicating these cells in many immune responses. Specific signals from the microenvironment in which mast cells reside can activate different molecular events that govern distinct mast cells responses. We previously demonstrated that hydrogen peroxide (H(2)O(2)) promotes IL-4 and IL-6 mRNA production and potentates FcepsilonRI-induced c...
متن کاملSuppression of Ribosomal Function Triggers Innate Immune Signaling through Activation of the NLRP3 Inflammasome
Some inflammatory stimuli trigger activation of the NLRP3 inflammasome by inducing efflux of cellular potassium. Loss of cellular potassium is known to potently suppress protein synthesis, leading us to test whether the inhibition of protein synthesis itself serves as an activating signal for the NLRP3 inflammasome. Murine bone marrow-derived macrophages, either primed by LPS or unprimed, were ...
متن کاملMast Cells Respond to Cell Injury through the Recognition of IL-33
Mast cells have been attributed several functions in both health and disease. Mast cell activation and release of inflammatory mediators are associated with the pathogenesis of several diseases, in particular that of allergic diseases. While the notion of mast cells as important, protective sentinel cells is old, this feature of the cell is not well recognized outside the mast cell field. The m...
متن کاملA2E Induces IL-1ß Production in Retinal Pigment Epithelial Cells via the NLRP3 Inflammasome
AIMS With ageing extracellular material is deposited in Bruch's membrane, as drusen. Lipofuscin is deposited in retinal pigment epithelial cells. Both of these changes are associated with age related macular degeneration, a disease now believed to involve chronic inflammation at the retinal-choroidal interface. We hypothesise that these molecules may act as danger signals, causing the productio...
متن کاملNotochordal cells protect nucleus pulposus cells from degradation and apoptosis: implications for the mechanisms of intervertebral disc degeneration
INTRODUCTION The relative resistance of non-chondrodystrophic (NCD) canines to degenerative disc disease (DDD) may be due to a combination of anabolic and anti-catabolic factors secreted by notochordal cells within the intervertebral disc (IVD) nucleus pulposus (NP). Factors known to induce DDD include interleukin-1 beta (IL-1ß) and/or Fas-Ligand (Fas-L). Therefore we evaluated the ability of n...
